Signalling pathways in oocyte meiotic maturation.

چکیده
Immature oocytes become fertilisable eggs through a process called meiotic maturation, which is often induced by specific hormones. Oocyte maturation involves the activation of various signal transduction pathways that converge to activate maturation-promoting factor (MPF); this is a key activity that catalyses entry into M-phase of meiosis I and meiosis II. Whereas the function of MPF in promoting oocyte maturation is ubiquitous, there are species-dependent differences in the signalling pathways leading to MPF activation. The poster is based mainly on the signalling pathways elucidated in oocytes of the African clawed frog Xenopus laevis, which is probably the most intensively studied model system for meiotic maturation. Important differences between oocytes from Xenopus and other organisms are discussed below. Further details and primary references can be found in several recent reviews (Abrieu et al., 2001; Ferrell, 1999; Karaiskou et al., 2001; Maller et al., 2001; Nebreda and Ferby, 2000; Yamashita et al., 2000). Fully grown oocytes are arrested at the first meiotic prophase. The hallmarks of meiotic maturation in vertebrate oocytes are (1) resumption of meiosis I which includes germinal vesicle breakdown (GVBD), chromosome condensation and spindle formation, (2) the transition between meiosis I and meiosis II, including inhibition of S-phase and (3) arrest in metaphase II because of cytostatic factor (CSF) activity. Meiosis II is completed after fertilisation of the mature oocyte (or egg). By contrast, oocytes of many invertebrates proceed during maturation only to metaphase of meiosis I, which is when they are fertilised.

P-106: Effect of Cilostamide on Meiotic Arrestof Ovine Oocytes
Background: in vitro maturation (IVM) of oocytes has increasing potential applications in assisted reproductive technology (ART), during which germinal vesicle (GV) oocytes cultured in vitro to produce mature (MII arrested) ones. Although functional, IVM oocytes have low developmental competence compared to in vivo matured oocytes; possibly because IVM comprises asynchrony between nuclear and c...
متن کاملP-131: Effects of Heat Shock during Early Stage of Oocyte Maturation on Meiotic Progression and Subsequent Embryonic Development and Gene Expressionin Ovine
Background: Heat shock may affect different aspects of oocyte maturation and its subsequent development to the blastocyst stage. A series of in vitro experiments was performed to determine whether physiologically heat shock (41°C) disrupts the progression of the ovine oocytes through meiosis, activation and blastocyst formation. Materials and Methods: The cumulus-oocyte complexes (COCs) were as...
متن کاملControl of oocyte growth and meiotic maturation in Caenorhabditis elegans.
In sexually reproducing animals, oocytes arrest at diplotene or diakinesis and resume meiosis (meiotic maturation) in response to hormones. Chromosome segregation errors in female meiosis I are the leading cause of human birth defects, and age-related changes in the hormonal environment of the ovary are a suggested cause. Caenorhabditis elegans is emerging as a genetic paradigm for studying hor...
متن کاملStops and starts in mammalian oocytes: recent advances in understanding the regulation of meiotic arrest and oocyte maturation.
Mammalian oocytes grow and undergo meiosis within ovarian follicles. Oocytes are arrested at the first meiotic prophase, held in meiotic arrest by the surrounding follicle cells until a surge of LH from the pituitary stimulates the immature oocyte to resume meiosis. Meiotic arrest depends on a high level of cAMP within the oocyte. This cAMP is generated by the oocyte, through the stimulation of...
متن کاملO-11: Zona Pellucida Birefringence and Meiotic Spindle Visualization of Human Oocytes Are Not Influenced by In Vitro Maturation Technology
Background: In vitro maturation (IVM) is a promising treatment option for certain infertile women. Nowadays, with the aid of the Polscope, it has become possible to evaluate zona pellucida (ZP) and spindle as parameters of oocyte quality. The goal was to investigate the relationship between the presence of the meiotic spindle and ZP birefringence with morphology of the in vivo and in vitro matu...
